摘 要: 目的探讨Wnt/β-catenin信号通路在哮喘大鼠气道平滑肌细胞中的表达及其对细胞迁移能力的影响。方法建立哮喘大鼠模型,原代培养大鼠气道平滑肌细胞(ASMC),采用反转录-聚合酶链反应(RT-PCR)、Western blot方法分别检测正常大鼠平滑肌细胞和ASMC中β-catenin信号通路mRNA和蛋白表达水平;采用Transwell迁移实验检测ASMCs的迁移能力,并分析应用β-catenin siRNA对其迁移能力的干预作用。结果 RT-PCR和Western blot结果显示哮喘大鼠ASMCs中β-catenin的mRNA和蛋白表达水平较对照组显著升高;β-catenin特异性siRNA能够降低ASMCs中β-catenin mRNA和蛋白表达水平;沉默β-catenin能够显著降低ASMCs的迁移能力。结论哮喘大鼠模型ASMCs的迁移能力显著增强,β-catenin信号通路的异常激活可能参与了这一生物学事件。 Objective To investigate the role of Wnt/β-catenin signaling pathway in migration ability of asthmatic rat airway smooth muscle cells (ASMCs). Methods The asthmatic rat airway smooth muscle cells were cultured in vitro. The expression of Wnt/β-eatenin in ASMCs was detected by RT-PCR and Western blot. Migration ability of ASMCs was detected by wound healing assay and transwell migration assay. Results Compared with normal rats,the expression level of Wnt/β-catenin in chronic asthmatic rats ASMCs was obvious higher. The β-eatenin signaling pathway by specific siRNA could reduce beta-catenin mRNA and protein expression level of ASMCs. The knockdown of Wnt/β-catenin was able to decreases the migration ability of ASMCs. Conclusion Abnormal activation of Wnt/β-catenin signaling pathway may participants in the migration ability of ASMCs of chronic asthma rats.
