摘 要: 探讨六堡茶总黄酮(LBTFE)对小鼠化学性肝损伤的保护作用及机制。给予肝损伤小鼠不同剂量LBTFE灌胃15 d后,测定体重并计算肝脏指数。检测血清肝功能相关指标酶(天门冬氨酸氨基转移酶、丙氨酸氨基转移酶、碱性磷酸酶、乳酸脱氢酶)和炎性因子(肿瘤坏死因子-α和白介素-6)水平。测定肝脏超氧化物歧化酶、谷胱甘肽、过氧化氢酶和丙二醛及离子通道酶活性。LBTFE能降低肝损伤小鼠血清肝功能相关指标酶、炎性因子和肝脏丙二醛水平。LBTFE还能增强抗氧化物酶,谷胱甘肽及离子通道酶的活力(p<0.05)。结果显示,LBTFE对小鼠化学性肝损伤有一定程度的保护作用,其机制可能与增强内源性抗氧化系统和调控离子通道酶的活性有关。 To study the protective effect of Liubao tea total flavonoid(LBTFE) on chemical induced liver injury mice, the treated mice were treated with different dose of LBTFE for 15 days. The body weight and liver index was measured at the end of experimental period. The serum levels of aspartate transaminase (AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), lactate dehydrogenase(LDH) and tumor necrosis factor (TNF)-α, interleukin (IL)-6 were determined by commercial assay kits. Hepatic levers of superoxide dismu-tase(SOD), catalase(CAT), glutathione(GSH), malondialdehyde(MDA), and Na+/K+-ATPase, Ca2+/Mg2+-ATPase were determined by commercial assay kits. LBTFE treatment reduced the serum levels of ALT, ALP, LDH, TNF-α, IL-6 and MDA in liver damage mice. LBTFE also increased the liver levels of SOD, CAT, GSH and Na+/K+-ATPase,Ca2+/Mg2+-ATPase in liver damage mice (p〈0.05). These results suggested that the LBTFE exhibited a protective effect to against the liver injury may associated with the increasing of endogenous antioxi-dant system and regulating the activity of Na+/K+-ATPase and Ca2+/Mg2+-ATPase in chemical induced liver damage mice.
