siRNA干扰白介素6抑制宫颈癌C33a细胞侵袭能力的研究
siRNA of IL-6 inhibits the invasion of C33a cells in cervical cancer

摘  要: 目的:探讨白细胞介素6(IL-6)对宫颈癌C33a细胞侵袭能力的影响。方法:宫颈癌C33a细胞分为对照组和siIL-6组(RNA干扰处理组),利用新型阳离子脂质体转染试剂Hilymax导入IL-6 RNA干扰序列,并检测IL-6基因的表达,而后应用Transwell实验检测C33a细胞侵袭能力的改变。结果:与对照组比较,qPCR实验结果显示,RNA干扰组IL-6 mRNA下降了41%(P<0.01),显示成功干扰了IL-6的基因表达;Transwell实验结果显示,较之于对照组,干扰组IL-6 C33a细胞侵袭细胞数减少(P<0.01)。结论:本研究成功干扰了IL-6基因表达,并且IL-6表达抑制导致C33a细胞侵袭能力下降。 Objective To investigate the influence of IL-6 on the invasion of cervical cancer cell, C33a.Methods C33a cells were divided into control group and siIL-6 group (RNA interference treatment group). IL-6 RNA interference sequences were introduced by a novel cationic liposome transfection reagent, Hilymax. IL-6 gene expression was detected through PCR, and C33a cell invasion was detected by Transwell experiment.Results Compared with the control group, qPCR showed that the IL-6 mRNA of silL-6 group was decreased by 41% (P 〈 0.05), indicating that this experiment successfully interfered with the gene expression of IL-6,Transwell experiment showed that compared with the control group, the cell invasion of the silL-6 group decreased significantly (P〈0.01).Condusion This study successfully interferes IL-6 expression and siIL-6 leads to the inhibition of cell invasion in C33a cells.
