IL-27及其受体在未足月胎膜早破发病中的相关研究

摘  要: 目的:探讨白介素-27(interleukin-27,IL-27)及其特异性受体WSX-1/TCCR诱导人胎膜组织产生炎性效应在未足月胎膜早破中的作用。方法:(1)将手术室收集到的胎膜组织分为正常足月组和未足月胎膜早破组,将培养的胎膜组织分为对照组、实验组(IL-27处理12 h组以及IL-27处理24 h组);(2)Western blot和免疫组化分析WSX-1在未足月胎膜早破早产和正常足月孕妇胎膜组织中的表达;(3)培养原代人胎膜组织,鉴定其是否存活;(4)IL-27处理培养的胎膜组织,提取处理的人胎膜组织蛋白,用荧光定量PCR和明胶酶谱检测MMP2、MMP9的表达。结果:(1)IL-27特异性受体WSX-1在未足月胎膜早破早产孕妇胎膜组织中的表达明显高于正常足月孕妇(P=0.0149);(2)成功培养原代人胎膜组织;(3)人重组IL-27蛋白刺激胎膜组织后MMP2、MMP9的表达明显升高。结论:IL-27可诱导人胎膜组织产生炎性效应,增强MMP2和MMP9的表达,参与未足月胎膜早破早产的发病。 Objective:To discuss the role of interleukin-27(IL-27)and its special receptor WSX-1/TCCR in inducing inflammatory effects on human fetal membrane in preterm premature rupture of membrane. Methods:(1)The expression of IL-27 special receptor WSX-1 in fetal membrane of preterm premature rupture of membrane and normal term birth was analyzed by western blot and immunohistochemistry assay.(2)Primary human fetal membrane was isolated and cultured and its survival was identified by RT-PCR.(3)The fetal membranes were divided into control group and the experimental group,treated by 10 ng/m L human recombinant IL-27 and then RNA and protein were extracted. MMP2 and MMP9 m RNA level was detected and their activity was examed by quantitative RT-PCR and Gelatin zymography,respectively. Results:(1)The expression of IL-27 receptor WSX-1 had significant differences between preterm premature rupture of membrane and normal term birth.(2)Primary human fetal membrane was successfully isolated and cultured for processing.(3)Expressions of MMP2 and MMP9 were obviously higher in IL-27 treatment group;P〈0.05 was statistically significant. Conclusion:IL-27 could induce human fetal membrane to produce MMP2 and MMP9 in the pathogenesis of preterm premature rupture of membrane.
