喀斯特地貌中碳酸酐酶微生物鉴定与特性研究

摘  要: 碳酸酐酶(CA)广泛分布于动植物和原核生物中,可以迅速催化CO2和HCO_3^-之间的相互转化反应,而喀斯特地貌独特的环境条件非常适合微生物碳酸酐酶的表达。选取具有典型喀斯特地貌的重庆金佛山景区的土壤样本,分离出高产碳酸酐酶菌,选取酶活性最高的一株菌,进行革兰氏染色镜检,测定生长曲线确定稳定期,最后对其进行16S rRNA序列分析,鉴定种属,同时探究了循环水系统的主要环境因子(温度、pH值、金属离子、阴离子)对其碳酸酐酶活性的影响。结果表明:筛选出酶活高达2.12 U/mg的菌株C8,其为革兰氏阳性菌,呈短杆状,于18 h达到生长稳定期,C8的16S rRNA序列分析的鉴定结果为Bacillus sp.;Bacillus sp.菌CA具有良好的热稳定性,中性弱碱性环境有利于CA活性相对稳定,金属阳离子Ca^(2+)、Mg^(2+)、Zn^(2+)、Fe^(2+)以及阴离SO_4^(2-)、Cl^-在一定范围内可使B.RA51菌CA活性保持相对稳定。此研究为高产微生物碳酸酐酶应用于循环冷却水系统提供了参考。 Carbonic anhydrase(CA) which can catalyze inter-conversion between CO_2 and HCO_3^- rapidly exists widely in prokaryote cells,and the unique conditions of Karst landform is very suitable for the expression of microbial carbonic anhydrase. In this paper,the soil samples were collected from the Jinfoshan scenic spot with the typical Karst landform in Chongqing,isolating bacterium with strains high-yielding carbonic anhydrase. A strain with the highest enzyme activity was selected to perform gram stain microscopy and evaluate the stationary phase. Getting the species of the strain by the identification result of 16 S rRNA. Then,it explores the influence of the main environmental factors(such as temperature,pH,mental ions and anions),on the activity of it 's carbonic anhydrase. It turned out that the bacterial strains C8 is a gram positive bacterium,which has a short pole shape and reaches the stable stage in 18 hours. And the identification result of 16 S rRNA is the Bacillus sp. The carbonic anhydrase activity of the Bacillus sp have good thermal stability. Neutral and alkaline of the environment was fit for keeping stability of CA. A certain range of concentration of mental ions such as Ca^(2+),Mg^(2+),Zn^(2+)and Fe^(2+)along with anions such as SO_4^(2-) and Cl^- have few effects on the CA activity of B. RA51. This research provides scientific basis for applying the B.RA51's CA in circulating cooling water.
