摘 要: 在5 L发酵罐上,以考察搅拌转速、补料及p H值对菌株Pseudoalteromonas carrageenovora ASY5产卡拉胶酶的影响进行优化,建立卡拉胶酶的高产发酵工艺。结果表明:提高搅拌转速能促进产酶并缩短发酵时间,其中搅拌转速为280 r/min时较佳;发酵过程中补料或控制恒定p H值对酶活力没有促进作用。在5 L罐发酵试验基础上进行中试放大实验,在20,75,200 L罐中的卡拉胶酶活力最大值分别为33.9,34.7,36.1 U/m L。 In the 5 L fermentor, the effects of rotation speed supplementation of medium and pH on the production of carrageenase by strain Pseudoalteromonas carrageenovora ASY5 were investigated to optimize fermentation process. The results showed that high rotation speed could promote the rate of enzyme producing and shorten the fermenting time, and a speed of 280 r/min was found to be the most appropriate. Additional supplementation of medium and control constant pH exerted no obvious effects on the yield of enzyme produc- tion. Then, the scale-up experiments of 20, 75 and 200 L were conducted based on the results of 5 L biore- actor. The maximum carrageenase activitives reached 33.9, 34. 7 and 36. 1 U/mL in 20, 75 and 200 L bio- reactor, respectively.
