摘  要: 目的建立丙酮/二乙醚溶液和水方法诱发的C57BL/6小鼠干皮症模型(AEW模型),检测胃泌素释放肽(GRP)及受体(GRPR)在颈段背根神经节(DRG)及脊髓的表达,并探讨它们在AEW诱发的干皮症慢性瘙痒发病机制中的作用。方法外用丙酮/二乙醚混合液(1∶1)和水7d建立C57BL/6小鼠的AEW模型,观察小鼠搔抓行为学,皮肤干燥症状,皮肤组织学(HE染色)和肥大细胞浸润(甲苯胺蓝染色)等指标,定量PCR分析GRP在颈部背根神经节(DRG),GRPR在颈部脊髓的mRNA表达水平变化。结果与对照组相比,AEW模型组小鼠的自发性搔抓次数明显多于对照组;AEW模型组小鼠颈背部皮肤表现出明显干燥、脱屑,病理提示表皮增厚、真皮肥大细胞浸润不明显;在颈部DRG的GRP、在颈部脊髓的GRPR mRNA表达水平升高。结论成功建立了干皮症且伴有强烈搔抓行为的C57BL/6小鼠模型,GRP和GRPR在AEW模型瘙痒中有重要作用。 Objective To establish a human dry skin disorder model in C57BL/6 mice, acetone/ether mixture followed by water (AEW) was applied and gastrin-releasing peptide(GRP) and receptor (GRPR) mRNA levels in the cervical DRGs and spinal cord were measured and their roles in pathogenesis of chronic itch caused by skin dryness were discussed. Methods C57BL/6 mice were treated daily with cutaneous application of acetone/ether ( 1 : 1 ) mixture (AE) and followed by water (AEW) for 7 days. The dermatitis symptoms, including mouse scratching behavior, dryness of the skin, skin histology (by HE staining) and mast cell infiltration (by toluidine blue staining) were evaluated. In particularly, mRNA levels of GRP and GRPR in dorsal root ganglia (DRGs) and spinal cord respectively, were detected by quantitative RT-PCR. Results The amount of spontaneous scratching of AEW model mice was increased significantly compared to the controis. AEW resulted in significant dryness and desquamation in skin, and thickening of epidermis indicated by the histology; however, did not alter degranulation of cutaneous mast cells. GRP and GRPR mRNA levels in cervical DRGs and spinal cord were markedly elevated by the AEW. Conclusion Topically applying AEW on the nape of neck skin of C57BL/6 mice could produce a reliable dry skin model in mice accompa- nied by intensive pruritus. GRP and GRPR played an important role in the pathogenesis of chronic itch in AEW induced dry skin diseases.
