广西不同地区显齿蛇葡萄茎中黄酮类成分含量测定
Determination of Contents of Dihydromyricetin and Myricetin in Ampelopsis Grossdentatae Stems from Different Areas of Guangxi

摘  要: 目的:建立显齿蛇葡萄茎中黄酮成分的含量测定方法,并分析广西不同地区显齿蛇葡萄茎中二氢杨梅素及杨梅素的含量。方法:采用高效液相色谱法,安捷伦Poroshell EC-C18(4.6mm×250.0mm,4.0μm)为色谱柱,以乙腈-0.1%磷酸为流动相,梯度洗脱,流速1mL·min-1,柱温25℃,检测波长0~23min,330nm;23~50min,375nm。结果:显齿蛇葡萄茎中二氢杨梅素和杨梅素进样量分别在0.800 5~9.606 0μg(r=0.999 87)和0.100~1.202μg(r=0.999 72)范围内呈良好的线性关系;平均加样回收率分别为98.1%、99.9%,RSD<3%,方法的稳定性、重复性和精密度均良好(RSD<3%)。桂林灵川和岑溪大隆的显齿蛇葡萄茎中二氢杨梅素和杨梅素的含量均较高,武鸣双江和金秀互助村的显齿蛇葡萄茎中二氢杨梅素和杨梅素含量均较低。结论:不同地区显齿蛇葡萄茎中二氢杨梅素和杨梅素的含量相差较大。所建立的方法可较好地去除杂质,分离效果好,测定结果准确性、重复性好。 Objective:To establish a method for determination of contents of flavonoids in Ampelopsis grossdentatae stems and ana- lyze determination of dihydromyricetin and myricetin in Ampelopsis grossdentatae stems from different areas of Guangxi. Methods: HPLC method was adoped. The seperation was performed on Agilent Poroshell EC-C18 (4.6mm× 250mm,4.0μm) column and the mobile phase consisted of acetonitrile and 0. 1% H3PO4 water solution in gradient elution at flow rate of lmL · min-1 , the column temperature was 25 ℃, the detection wavelengths were set at 330nm for dihydromyricetin and 375 nm for myrieetin. Results:The lin- ear range was 0. 8005~9. 606μg(r=0. 999 87)for dihydromyricetin and 0. 100~1. 202μg(r=0. 99972)for myricetin with average re- covery of 98.1 %and 99.9 % ,RSD〈3%, and the stability , repeatability and precision of the method were all better in all cases(RSD〈3 % ). The determined results showed that the contents of dihydromyricetin and myricetin in Ampelopsis grossdentatae stems from Ling Chuan of Gui Lin and Da Long of Cen Xi were higher , and the contents in Ampelopsis grossdentatae stems from Shuang Jiang of Wu Ming and Hu Zhu Cun of Jin Xiu were lower. Conelusion: The contents of dihydromyricetin and myricetin in Ampelopsis grossdentatae stems were quite different. The established method can be better to remove impurities,good separation,and the meas- ured results showed the high accuracy and good reproducibility.
