一测多评法测定藏药五味黄连丸中4种生物碱的含量

摘  要: 目的建立藏药五味黄连丸(黄连、红花、诃子、渣驯膏和麝香)中表小檗碱、黄连碱、巴马汀和小檗碱的一测多评含量测定方法。方法采用RP-HPLC法,以乙腈-水(1mL·L-1三乙胺和1mL·L-1甲酸)(30∶70)为流动相;检测波长:345nm;柱温:25℃;流速:0.8mL·min-1;以盐酸小檗碱为内标物,测定小檗碱、表小檗碱、黄连碱和巴马汀的相对校正因子,计算五味黄连丸中4种生物碱的含量,并比较一测多评法的计算值与常规外标法的实测值的相似度。结果一测多评法与常规外标法测得的结果差异无统计学意义,2组数据的夹角余弦值、相关系数和相似度均达到0.999。结论利用一测多评法可简便可靠、快速、准确地控制五味黄连丸的质量。 To estahlish a method for the simultaneous determination of epiberberine,coptisine,palmatine and berberine inTibetan medicine of Wuweihuanglian Pills ( Coptidis rhizoma , Crarthamus , myrobalan, trogopterus dung cream andmusk) by quantitatively analyzing multi-components with a single-marker (QAMS). RP-HPLC method was adopted. Themobile phase was acetonitrile-water (1 mL · L 1 triethylamine and 1 mL ·L 1 formic acid) (30 : 70),the flow rate was 0. 8 mL* min 1 with UV detection wavelength at 345 nm and the column temperature was maintained at 25 °C. With berberine hydro-chloride as an internal standard, the relative correlation factors (RCF) of epiberberine, coptisine and palmatine to berberine were determined. The content of these 4 alkaloids were determined by external standard method and QAMS,respectively. The similarity of those data was compared. No significant difference in assay results was observed. The included angle cosine, correlationand similarity all reached 0. 999. It is simple,reliable,rapid and accurate to evaluate the quality of Wuweihuanglian Pillsby QAMS.
