摘 要: 恶性肿瘤是世界大多数国家最主要的死亡原因之一,其发病率及死亡率在我国总体呈上升趋势。恶性肿瘤已严重影响人类健康,给社会和家庭带来沉重的经济负担。目前恶性肿瘤的发病机制尚不明确,治疗效果总体欠佳,治疗的靶向药物相对较局限,仍需进一步研究。DHX9(DEAH-box helicase 9)是一种NTP依赖的RNA/DNA解旋酶。作为一种多功能蛋白,DHX9能与多种分子伴侣相互作用,参与调节多个生物学过程,修复DNA损伤及维持基因的稳定。最近研究表明DHX9对多种肿瘤起促进作用,抑制DHX9的表达可对肿瘤细胞造成致命性伤害,但不影响正常细胞功能,提示DHX9可能成为恶性肿瘤治疗的一个潜在靶点。本文对DHX9对不同肿瘤发生发展作用的研究进行了综述。 Cancer is still one of the leading cause of death in most countries worldwide,and its incidence and mortality continuously increase in China. Cancer has heavily influenced our health,and constitutes an enormous burden on whole society and families. Until now,the pathogenesis of cancer is unclear. Meanwhile,overall therapeutic effect is poor,and the targeted drugs for cancer are relatively limited. Therefore,it is still necessary to identify more targets for cancer treatment. DHX9 is an NTP-dependent RNA/DNA helicases,and is widely expressed on various tissues and cells. The presence of numerous functional domains likely contributes to the multifunctional nature of DHX9,which has been implicated in a variety of biological processes. DHX9 is also an important factor in DNA repair and maintenance of genome stability,and is able to unwind aberrant polynucleotide structures. Recent studies indicate that DHX9 promotes the development of various tumors. The suppression of DHX9 expression can be lethal to tumor cells,yet it does not affect the function of normal cells,suggesting that DHX9 can be a potential targeted drug for cancer. This review will summarize the implication of DHX9 in various cancers.
