摘 要: 目的观察磷酸西格列汀对2型糖尿病(T2DM)大鼠肝脏组织亲吻素1(KISS1)水平的影响。方法 20只大鼠高脂饮食加小剂量链脲佐菌素诱导构建T2DM大鼠模型后均分为两组:实验(SIT)组实施磷酸西格列汀10mg·kg^(-1)·d^(-1)灌胃,模型(DM)组给予等体积生理盐水灌胃。另取10只大鼠(NC组)常规喂养,等体积生理盐水灌胃。干预12周后,采用qRT-PCR和Western blot分别检测各组大鼠肝组织KISS1 mRNA水平和蛋白表达。结果与NC组相比,DM组大鼠KISS1 mRNA和蛋白水平提高(P<0.05)。SIT组大鼠KISS1 mRNA和蛋白水平低于DM组(P<0.05)。结论磷酸西格列汀可有效降低T2DM大鼠肝脏组织KISS1水平的升高。 Objective To observe the effect of of sitagliptin on kisspeptin 1(KISS1)in the liver of rats with type 2diabetes mellitus(T2DM).Methods T2DM models were established by feeding high-fat diet and low-dose streptozotocin in 20 SD rats,which were equally assigned into two groups of A(treated with sitagliptin 10mg·kg-1·d-1 by gavage)and B(normal saline gavage).Another 10 SD rats were taken as blank controls(group C).The qRT-PCR and Western blot were used to detect the expressions of KISS1 mRNA and protein in liver tissues of rats after 12 weeks of intervention.Results The expressions of KISS1 mRNA and protein were significantly higher in group B than those in groups of C and A(P〈0.05).Conclusion Sitagliptin can effectively attenuate the increase of KISS1 in the liver of rats with T2DM.
