豆渣固定化培养乳酸菌的条件优化及其对乳酸菌的保护作用

摘  要: 以豆渣为乳酸菌固定化载体,利用单因素实验和正交实验对影响乳酸菌固定化培养的3个因素进行了优化,同时研究了体外模拟消化系统胁迫对发酵豆乳中活乳酸菌数量的影响。结果表明:固液比1∶40、接种量3%、培养时间24 h为豆渣固定化培养乳酸菌的最佳条件,在此组合条件下豆渣载体上固定吸附的最大活菌数为1.07×1010cfu/g;经体外模拟胃肠消化系统后,豆渣固定化乳酸菌发酵豆乳中的活菌数显著高于游离乳酸菌发酵豆乳中的活菌数。说明豆渣可以作为一种新型固定化乳酸菌的材料,且能够增强乳酸菌对消化系统胁迫的抵抗能力。 Soybean dreg(okara) was used as the carrier for the immobilized culture of lactic acid bacteria(LAB),three factors affecting the immobilized culture of LAB were optimized through single-factor test and orthogonal experiment,and the effects of simulated in-vitro digestion system stress on the quantity of live LAB in fermented soymilk were also studied. The optimum conditions for the immobilized culture of LAB on okara were obtained as follows: solid-liquid ratio 1 ∶ 40,inoculum size 3%,and culture time 24 h. Under these optimum conditions,the maximum quantity of live LAB immobilized and adsorbed by okara was 1.07× 1010cfu/g. Meanwhile,under the stress of simulated in-vitro stomach and intestine digestion system,the quantity of live LAB in the fermented soymilk inoculated with okara-immobilized LAB was significantly higher than that in the fermented soymilk inoculated with free LAB. The above results indicate that okara can be used as a new-type material to immobilize LAB,and it can strengthen the resistance of LAB to digestion system stress.
