摘 要: 目的:通过运用益气活血法的代表方剂补阳还五汤对动脉粥样硬化模型的干预,观察主动脉组织Rho激酶、VCAM-1及ICAM-1 mRNA的表达、血脂等指标的变化,以揭示该方药抗动脉粥样硬化作用的新靶点和新途径。方法:维生素D3加高脂饮食诱导大鼠动脉粥样硬化模型。60只大鼠用SPSS 13.0软件随机分为6组:正常组、模型组、补阳还五汤低剂量治疗组、补阳还五汤高剂量治疗组、辛伐他汀对照组、补阳还五汤预防组。造模成功后干预28 d,检测主动脉Rho激酶、VCAM-1及ICAM-1mRNA表达及血脂水平。结果:高脂饮食加维生素D3可诱导大鼠形成动脉粥样硬化模型,模型组有动脉粥样硬化斑块形成,补阳还五汤高剂量治疗组、辛伐他汀对照组、补阳还五汤预防组动脉粥样硬化病变程度明显减轻。Rho激酶、VCAM-1及ICAM-1mRNA表达量水平及血脂水平:模型组与正常组相比,有显著的统计学差异(P<0.01)。补阳还五汤低剂量治疗组与模型组相比有统计学意义(P<0.05),补阳还五汤高剂量治疗组、辛伐他汀对照组、补阳还五汤预防组与模型组相比均有显著的统计学意义(P<0.01),各干预组与正常组相比,有统计学差异(P<0.05)。补阳还五汤预防组、补阳还五汤高剂量治疗组、辛伐他汀对照组各组间无统计学意义(P>0.05)。结论:补阳还五汤可以下调Rho激酶、VCAM-1及ICAM-1mRNA的表达水平、降低血脂,具有抗动脉粥样硬化作用,降低Rho激酶、VCAM-1及ICAM-1mRNA水平可能是其作用机制之一。 Objective: We study the influence of Qi-benifiting and blood-activating representative formula Buyang Huanwu Decoction( BYHWD) on atherosclerosis model aorta Rho kinase,VCAM-1 and ICAM-1mRNA and the level of blood-lipid in order to reveal new targets and new ways of the anti-atherosclerotic role of the formula. Method: Vitamin D3 and high fat diet induced the atherosclerosis in rats. Sixty rats were divided into 6 groups by SPSS13. 0 software: normal control group,model group,low dose of BYHWD therapy group,high dose of BYHWD therapy group,simvastatin control group,BYHWD prevention group. After model successing and then 28 days therapy,detect the expression levels of Rho kinase,VCAM-1 and ICAM-1mRNA in atherosclerosis aorta by reverse trascription polymerase chain reaction( RT-PCR) and the level of blood-lipid. Result: High fat diet and Vitamin D3 could induce atherosclerosis in rats. Comparing with normal group,there was atherosclerotic plaque in model group,AS pathological changes less in high dose of BYHWD therapy group,simvastatin control group and BYHWD prevention group. Comparing with model group,the expressions of Rho kinase,VCAM-1 and ICAM-1mRNA and the level of blood-lipid of normal control group,high dose of BYHWD therapy group,BYHWD prevention group,simvastatin control group were decreased significantly( P〈0. 01).Those of low dose of BYHWD therapy group was decreased( P〈0. 05) but exceeded those in normal group( P〈0. 05).There was no statistically significant difference between the BYHWD prevention group,high dose of BYHWD therapy group,simvastatin control group( P〉0. 05). Conclusion: Buyang Huanwu Decoction has the effect of down-regulating the expressions of Rho kinase,VCAM-1 and ICAM-1mRNA and the level of blood-lipid in rat model. It has the effect of against atherosclerosis. Down-regulating the expressions of Rho kinase,VCAM-1 and ICAM-1mRNA may be one of the mechanisms of againsting atherosclerosis.
