摘 要: 目的探讨镇肝熄风汤对原发性高血压大鼠模型主动脉纤维化的治疗作用。方法以SHR大鼠为实验模型,以卡托普利为阳性对照药,观察镇肝熄风汤对原发性高血压的降压作用以及检测大鼠主动脉中TGF-β1、Ⅰ型胶原、CTGF基因相对表达量与Smad2蛋白表达。结果造模阶段与空白组比较各组血压均升高,差异有统计学意义(P<0.05),给药阶段与模型组相比各用药组血压均降低,差异有统计学意义(P<0.01)。与模型组比较,镇肝熄风汤和卡托普利均能降低血压并抑制主动脉中TGF-β1、Ⅰ型胶原、CTGF基因相对表达量,并降低Smad2蛋白的表达。结论镇肝熄风汤可降低大鼠血压并抑制主动脉纤维化,其作用可能是通过抑制主动脉中TGF-β1、Ⅰ型胶原、CTGF基因相对表达量,并降低Smad2蛋白的表达来实现的。 Objective To investigate the therapeutic effect of Zhenganxifeng decoction( liver settling-wind extinguishing decoction) on aortic fibrosis in rats with primary hypertension. Method SHR rats were used as experimental models,with Captopril as the positive control medicine. The antihypertensive effect of Zhenganxifeng decoction on of primary hypertension were observed and the relative expression of TGF-β1,collagen Ⅰ,CTGF gene in rat aortic and Smad2 protein expression were detected. Results Modeling stage was compared with control group,the blood pressure of each group were increased,and the difference was statistically significant( P〈0. 05). Compared with the model group,the blood pressure decreased in each medication group,and the difference was statistically significant( P〈0. 01). Compared with the model group,Zhenganxifeng decoction and captopril all can lower the blood pressure,and inhibit the relative expression of TGF-β1,collagen and CTGF gene in the aorta and reduce the expression of Smad2 protein. Conclusion Zhenganxifeng decoction can decrease the blood pressure of rats and inhibit aortic fibrosis,the effect may be achieved by inhibiting aortic TGF-β1,collagen and CTGF gene and decreasing the expression of Smad2.
