蒙医三根平衡针对慢性应激抑郁模型大鼠海马神经元形态及BDNF-CREB信号通路的影响

摘  要: 目的:观察蒙医三根平衡针对慢性应激抑郁模型大鼠海马神经元形态及BDNF-CREB信号通路的影响,以期探讨蒙医三根平衡针抗抑郁作用的机制。方法:雄性SD大鼠随机分为空白组、模型组、药物组、蒙医银针组、蒙医三根平衡针组和针药结合组,每组8只。除空白组外,每组大鼠孤养,进行不可预知的应激刺激,并接受不同的治疗。28d后检测行为学改变;断头、取材,观察CA3区神经元形态;检测海马BDNF、CREB的表达。结果:与空白组比较,模型组大鼠水平及垂直运动次数明显减少(P<0.05)。与模型组比较,蒙医三根平衡针组、药物组及针药结合组水平及垂直运动次数明显增加(P<0.05),海马CA3区神经元锥体细胞排列较有序、神经元胞膜较完整;海马BNDF、CREB的表达升高(P<0.05)。结论:蒙医三根平衡针治疗抑郁症的作用机制,与神经元保护及上调海马BDNF-CREB表达有关。 Objective: To observe the effect of Mongolian acupuncture ‘three modulation methods' on neurons form and BDNF-CREB signaling pathways in hippocampus in rats with chronic stress depression. To explore the antidepressant mechanisms for Mongolian acupuncture ‘three modulation methods' treatment. Methods: The male SD rats were randomly divided into the control, the model, the fluoxetine, the Mongolian acupuncture, the Mongolian acupuncture ‘three modulation methods', and the combination of Mongolian acupuncture ‘three modulation methods' with fluoxetine groups, 8 rats in each group. Rats model of depression were established by chronic unpredictable stress stimulation in combination with 28 days of solitary confinement in all groups except normal control group. Rats were received with different treatments. The behavior changes were observed after 28 days. Hippocampus were collected and observed of neurons form in CA3 area. The expression of BDNF, CREB was detected by Western blot. Results: Compared with the control group, crossing-movement times and rearing-movement times in model group were significantly decreased(P<0.05); compared with the model group, crossing-movement times and rearing-movement times in the Mongolian acupuncture ‘three modulation methods' group, the fluoxetine group, combination of the Mongolian acupuncture ‘three modulation methods' with fluoxetine group were significantly increased(P<0.05); neuronal pyramidal cells in hippocampal CA3 area arranged orderly and the nerve cell membrane was complete; BNDF and CREB protein expression were significantly increased(P<0.05). Conclusion: The antidepressant mechanisms of Mongolian acupuncture ‘three modulation methods' treatment are related to the neuroprotection and the up-regulation of BDNF-CREB expression.
