超大孔聚合物微球的亲水改性及在快速生化分离中的应用

摘  要:   HYDROPHILIC GIGAPOROUS POLYSTYRENE MICROSPHERES WERE FABRICATED BY SURFACE GRAFTING POLYMERIZATION OF METHACRYLOXYETHYLTRIMETHYL AMMONIUM CHLORIDE (DMC) USING ATOM TRANSFER RADICAL POLYMERIZATION (ATRP).IN THIS METHOD, GRAFTING AMOUNT OF PDMC WAS CONTROLLED THROUGH REACTION TIME AND THE TOTAL POLYMERIZATION TIME WAS OPTIMIZED FOR 2 H.THE MODIFIED MICROSPHERES COULD EXHIBIT A PROTEINS RECOVERY OF OVER 90%.THE MORPHOLOGY OF THE MODIFIED MICROSPHERES WAS OBSERVED BY THE SCANNING ELECTRONIC MICROSCOPE (SEM), AND SEM IMAGES INDICATED THE THROUGHPUT PORES IN THE GIGAPOROUS POLYSTYRENE WAS NOT BLOCKED BY THE GRAFTING POLYMERS.THE DYNAMIC BINDING CAPACITY OF PROTEINS ON THE MODIFIED MICROSPHERES WAS MEASURED IN VARIOUS FLOW RATES, AND THE RESULTS INDICATED THAT DBC WAS NOT OBVIOUS CHANGED AT THE RANGE OF 23-19 MG/ML WHEN THE FLOW RATES CHANGED FROM 360 CM/H TO 1800 CM/H.THE MODIFIED GIGAPOROUS MICROSPHERES WITH GOOD PERMEABILITY SHOWED GREAT POTENTIAL IN THE RAPID SEPARATION OF PROTEINS.
