VB/_1、VB/_(12)和VH对雨生红球藻(Haematococcus pluvialis)的生长及虾青素含量的影响

摘  要: 雨生红球藻/(Haematococcus pluvialis/)是绿藻门/(Chlorophata/)、绿藻纲/(Chlorophyceae/)、团藻目/(Volvocales/)、红球藻科/(Haematococcaceae/)、红球藻属/(Haematococcus/)的单细胞经济微藻,含有大量虾青素,可用作饲料添加剂、天然食用色素和保健品、药品原料等。为了探讨VB/_1、VB/_/(12/)和VH对雨生红球藻的生长及虾青素含量的影响,设置了维生素B/_1、维生素B/_/(12/)和维生素H三种维生素单独添加、两种和三种组合添加的七组试验。测定了细胞密度、生物量、叶绿素a含量、虾青素含量等指标。结果表明:单独添加维生素B/_1、B/_/(12/)和H均对雨生红球藻游动细胞的生长有明显的促进作用。两种或三种同时添加,雨生红球藻的细胞密度、生物量和叶绿素a含量等指标均随着添加浓度的增加而上升。到达最高点后随着浓度的升高开始降低。在最高浓度下各指标均低于对照。VB/_/(12/)的促进效果优于其它添加处理,其添加浓度50μg·L~/(-1/)时,细胞密度、生物量和叶绿素a含量均显著高于其它处理,分别比对照提高了29.5%、30.0%和28.0%。单独添加维生素B/_1、B/_/(12/)和H能够促进雨生红球藻虾青素含量增加。两种或三种同时添加时,在低浓度时也对虾青素有促进作用。在所有组合及浓度处理中,VB/_/(12/)添加浓度为50μg·L~/(-1/)时,藻液虾青素含量为最高/(26.04μg·mL~/(-1/)/),比对照提高了23.65%。在雨生红球藻及其虾青素规模化生产过程中,可以通过适当地添加VB/_/(12/)来达到提高藻细胞密度、生物量、叶绿素a和虾青素含量的目的。 Haematococcus pluvialis that belongs to Chlorophata, Chlorophyceae,Volvocales, Haematococcaceae is a single economic microalgae and accumulates remarkable amounts of natural astaxanthin, which is an excellent feed additive, food colorant and a potential medicine. In order to grope for effect of Vitamin-B/_1, Vitamin-B/_/(12/) and Vitamin-H on the growth and astaxanthin content of Haematococcus pluvialis. The experiments of Adding Vitamin-B/_1, Vitamin-B/_/(12/) and Vitamin-H are setup,and is conducted for adding respectively, adding two kinds together and adding three kinds together. The experiments is up to seven groups. Cell density, biomass, chlorophyll-a content were measured. The results indicated that: The growth of H. pluvialis was improved significantly by adding of Vitamin-B/_1, Vitamin-B/_/(12/) and Vitamin-H respectively, together two kinds or together three kinds. The cell density, biomass, chlorophyll-a content would obviously enhance with the increase of concentration of every group/(except for the high concentration of every group/). The growth of H. pluvialis in the Vitamin-B/_/(12/) group was better than other groups, and under the 50μg·L~/(-1/) concentration of Vitamin-B/_/(12/), cell density, biomass, chlorophyll-a content were highest in all treatments and enhanced with 29.5/%, 30.0/%, 28.0/% higher than the control. Astaxanthin content of H. pluvialis was accelerated significantly by adding of Vitamin-B/_1, Vitamin-B/_/(12/) and Vitamin-H. when adding concentration of Vitamin-B/_/(12/) was 50μg·L~/(-1/) astaxanthin content was highest/(28.35μg·mL~/(-1/)/), enhanced with 28.31 /% than the control. Cell density and astaxanthin content were corresponding relation, properly adding Vitamin not only accelerated Cell density significantly but also enhance astaxanthin content. On the mass culture of motile cells of Haematococcus pluvialis properly adding Vitamin-B/_/(12/) was effective for increase cells density, biomass, chlorophyll-a and astaxanthin content.
