荧光共轭寡聚体的合成、自组装及应用研究

摘  要: 共轭寡聚体具有丰富的物理化学性质,它的结构繁多,易于调控。其构建的纳米材料由于形貌规整,功能易于设计,可应用于生物体系等优点受到人们广泛的关注,在电子器件、细胞成像、检测传感等领域有着重要的应用。在检测传感的方法中,荧光探针法是目前该领域热门的研究方法,荧光探针能够对特定的被检物种进行微量、快速、低成本检测,已经广泛应用于环境监测、疾病诊断、食品安全等许多领域。 基于以上两点制备了一种荧光纳米粒子,主要包括两部分内容: 在第一部分工作中,利用Sonogashira偶联反应,合成了两种水溶性良好的共轭寡聚体,它们在水中可以自发的形成球状的纳米粒子,对其光谱、形貌进行了表征。细胞成像实验表明,此纳米粒子有着良好的生物安全性,能够很好地被细胞吸收,并会聚集在细胞质区域。 在第二部分工作中,通过自组装方法成功的将制得的纳米粒子功能化,并对其光谱、形貌进行了表征。成功的实现了检测,且识别迅速。 Conjugated oligomers have abundant physicochemical property, which are complicatedstructured and easy controlled. Because the nano-materials, which are constituted byconjugated oligomers, have so many advantages, such as, well-shaped surface, easy designedfunction, and applicable using in biological systems that have been paid attention widely andthose also have been used in the fields of electron devices, cell imaging, and testing-sensorsignificantly. About testing-sensor, fluorescence probe technique is one of the hottest researchmethods of this field recently. Fluorescent probe is widely applicable in various fields,including analytical and environmental chemistry, clinical diagnostics, food industry, etc. dueto their simplicity and high sensitivity Based on the above two points we made a kind of fluorescent nanoparticles, there aremainly two parts in the thesis: In the first part of this thesis, based on Sonogashira coupling reaction mechanism,wesynthesizd two kinds of water-soluble conjugated oligomer. The self-assembled oligomer inwater were nanoscale spheres observed by transmission electron microscope.The opticalspectrum and particle morphology of nanospheres were characterized.Cell imagingexperiments showed that the nanospheres have good biological safety,can be absorbedefficiently by the cell and cumulated in the cytoplasm. In the second part of the thesis, by self-assembly method the functional nanoparticleswere successfully perpared.As well the optical spectrum and particle morphology ofnanospheres were characterized. The fluorescent probe of nano-particles can detect quickly.
