扶芳藤多糖的提取、分离、纯化及结构分析

摘  要: 扶芳藤属于卫矛属,是一种生长范围较广的藤条绿化植物,四处攀援生长,叶质柔软,叶子薄且呈椭圆形。扶芳藤也是一种中药材。在民间扶芳藤有岩青藤、万年青、换骨筋等多种称呼,其茎叶可以入药。据《本草拾遗》载,扶芳藤“味苦,小温,无毒”。功能与主治:舒筋活络,止血消瘀。治疗腰肌劳损,风湿痹痛,咯血,血崩,月经不调,跌打骨折,创伤出血。 目前以扶芳藤茎叶为原料提取分离其小分子化学成分的研究取得了一些研究成果,对扶芳藤粗多糖的及生理活性研究发现粗多糖提取分离具有提高免疫功能的作用。 但目前对扶芳藤多糖进行纯化分级、结构分析未见文献报道。 本论文分为两部分,第一部分对扶芳藤的化学成分、生物活性研究进展,以及多糖的提取分离方法,构效关系研究进行了综述。第二部分详述了论文实验过程:扶芳藤多糖的提取分离、纯化分级和结构分析。 经过石油醚、乙醚除脂去杂的扶芳藤茎叶粗粉,用热水浸取、过滤、浓缩、醇析、干燥后,得到深褐色的扶芳藤粗多糖。粗多糖经过脱蛋白和透析后,用Sephadex-75凝胶柱层析纯化得到一种纯化的多糖P/_1;用凝胶色谱法和比旋光度法鉴定多糖P/_1的纯度后,用红外光谱对其结构做了表征,使用乌氏粘度法测定了特性粘数,计算出了多糖的分子量。 将多糖P/_1完全酸水解后,通过高效液相色谱法确定了多糖的组成。其中多糖P/_1由6种单糖组成;其中能够确认的有甘露糖、鼠李糖、葡萄糖、阿拉伯糖;通过质谱分析,可以推断的有半乳糖、半乳糖醛酸。通过~1HNMR分析,进一步推断有甘露糖、葡萄糖、鼠李糖、半乳糖、阿拉伯糖;通过高碘酸氧化,可以确定多糖链主要连接方式为1→4连接;Smith降解后,发现多糖的主要连接方式为1→4连接,有少量的1→6连接,而没有1→3连接;通过~/(13/)CNMR分析,不仅确证了多糖P/_1的连接方式有α1→4连接,而且确证了有甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖存在。 根据上述实验结果和波谱分析,可以推断:扶芳藤多糖P/_1是由甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖构成,主要糖苷键构型为α-1→4连接,其次是1→6连接,没有1→2和1→3连接。 The wintercreeper euonymus is one kind of growth scope broad afforestation plant which belongs to the euonymus alata, it has green rattan, climbs up the growth, the leaf is soft thin and elliptic, Simultaneously the wintercreeper euonymus is also one kind of traditional Chinese medicine which is called as Yangqingteng、Wannianqing、Huangujin in the folk. Its stem and leaf may be taken raw material for medicine. According to "Book on Chinese medicine Appropriates lost articles" carries, common wintercreeper euonymus“taste pain, small warm, non-toxic”. Function and treatment: Relaxes the meridians and collaterals,treatment strain of lumbar muscles, rheumatism pain, hemoptysis, blood from uterus, irregular menstruation, dislocaitons and muscles injuries ,various fractures, bleeding trauma. At present some research results have been obtained in extracting small molecular chemical ingredient as raw material in stem and leaf of the common wintercreeper euonymus. The common wintercreeper euonymus thick raw polysaccharide also has been carried on the extraction to separate and discoved that it can enhane the immunologic function. Now any literature report has not been seen about the purification graduation, the structure analysis and oxidation resistance activity and the modified research to the common wintercreeper euonymus polysaccharide. This paper is written as two parts. The first part is a review, it carries on the summary to the common wintercreeper euonymus and the polysaccharide, introducing the common wintercreeper euonymus chemical composition and the polysaccharide pharmacological action, in addition introducing the polysaccharide extraction separation, purification graduation, structure analysis and medicine survey active. The second part in detail relates the paper experiment, mainly has written the common wintercreeper euonymus polysaccharide extraction separation, the purification graduation, the structure analysis. After the benzine, the ether excepted the fat and the impurity, and after the hot water leaching、the alcohol precipitation、the concentration、drying, it might be obtained the dark brown thick polysaccharide from the common wintercreeper euonymus stem leaf cribble. After the thick polysaccharide passed through deproteinizing and dialysis, it could be obtained one kind of purification polysaccharide P/_1 by the Sephadex-75 gelatin column chromatographic purification; After the purity was identified by the gel permeation chromatography and the specific rotation, P/_1 structure was characterized by FTIR spectra,and its molecular weight was calculated correspondingly by the intrinsic stickiness digital obtained with an Ubbelohde viscometer. After complete acid hydrolysis, polysaccharide composition was determined through HPLC, and polysaccharide P/_1 was composed of six kind of monosaccharides,and they could be confirmed were mannose, rhamnose, glucose,arabinose. Through the mass spectrum analysis, they might be infered there were galactose and galacturonic acid; Through the ~1HNMR analysis, they may be infered further that they are mannose, rhamnose, glucose,galactose, arabinose; Through periodate oxidation the polysaccharide chain main connection mode might be determined for 1→4 connection;After Smith degeneration, it was discovered that there was partly the 1→6 connection, but not the 1→3 connection in the polysaccharide; Through the ~/(13/)CNMR analysis, it was not only irrevocably proven the polysaccharide P/_1 connection mode was Alpha 1→4 connection, but also were they irrevocably proven that there were mannose, rhamnose, galacturonic acid, glucose , galactose, arabinose. According to the experimental result and the spectrum analysis, Common Wintercreeper Euonymus Polysaccharide P/_1 may be infered that it is compased of mannose, rhamnose, galacturonic acid, glucose,galactose and arabinose, as well as 6 kind of constitutions. The main connection in P/_1 isα-1→4, the next is 1→6 connection, no the collection of 1→2 and 1→3 the connection is observed.
