摘 要: 应用无特定病原体 (SPF)鸡胚增殖禽流感病毒 A/ Turkey/ Ontario/ 6 118/ 6 8(H8N4 )毒株 ,Tri Zol L S Reagent提取病毒 RNA,RT- PCR扩增神经氨酸酶 (NA)基因全片段 ,克隆到 p MD18- T载体上 ,并进行了鉴定和序列测定。所获得的 NA基因片段长 14 4 1bp,编码 4 90个氨基酸残基。根据推导的氨基酸序列进行预测 ,有 9个潜在的糖基化位点和2 Avian influenza viruses(AIV) are divided into different subtypes based on the antigenicity of the surface protein hemagglutinin(HA) and Neuraminidase(NA),and 15 HA subtypes and 9 NA subtypes have been isolated from birds.The influenza viruses with N1(H1N1,H5N1) or N2(H2N2,H3N2,H5N2 and H9N2) are widely circulated in birds,pigs and humans,and influenza viruses with N6(H4N6) and N8(H3N8) are circulated in pigs and horses,respectively,however,the influenza viruses with other NA subtypes are only seldom isolated from wild birds,and the sequence information of these NAs are very limited in the GenBank.In this study,the NA gene of N4 subtype influenza virus A/Turkey/Ontario/6118/68(H8N4) was amplified,cloned and sequenced.Results showed that the N4 NA gene has one open reading frame and encodes 470 amino acids.Deduced amino acids sequence showed that there are 9 potential glycosylation sites and 20 cysteines in the N4 NA protein.The sequence in this study is the first report of the full length N4 NA gene,and it will provide the necessary information for the further analysis of the structure and function of N4 NA of AIV.
