摘 要: 目的 :研究蛇毒Ⅱ类磷脂酶A2 (PLA2 )中D4 9PLA2 和K4 9PLA2 的功能分化及其功能分化决定位点的鉴定。方法 :运用序列比较分析 ,进化树构建和DIVERGEv1 0 4软件计算研究D4 9PLA2 和K4 9PLA2 的功能分化情况及其分化位点。结果 :序列比较分析 ,进化树构建和DIVERGEv1 0 4软件计算结果表明蛇毒Ⅱ类PLA2 中D4 9PLA2 和K4 9PLA2 的确发生了功能分化 ,对于K4 9PLA2 来说 ,1S ,7K ,11Q ,E12 ,R34,T5 6 ,N88,L92 ,E10 8,K116 ,K12 8可能为功能分化决定位点。对于D4 9PLA2 ,L2 ,G33,G35 ,F4 6和Y118可能为功能分化决定位点。结论 :我们首次通过序列比较分析 ,进化树构建和DI VERGEv1 0 4软件计算鉴定出蛇毒Ⅱ类PLA2 中D4 9PLA2 和K4 9PLA2 可能的功能分化位点 ,为今后通过基因重组和定点突变方法研究蛇毒Ⅱ类PLA2 Objective:To study the functional divergence of snake venom group Ⅱ phospholipase A 2(PLA 2)and corresponding sites in both D49 PLA 2 and K49 PLA 2.Methods:Multiple alignment,phylogenetic analysis and functional divergence analysis were employed.Results:Functional divergence existed in D49 PLA 2 and K49 PLA 2.The sites for functional divergence were 1S,7K,11Q,E12,R34,T56,N88,L92,E108,K116,K128 in the K49 PLA 2 and L2,G33,G35,F46 and Y118 in the D49 PLA 2.Conclusion:It is the investigation on the functional divergence of snake venom D49 PLA 2 and K49 PLA 2 with a combination of multiple alignment,phylogenetic analysis and functional divergence analysis methods.The identification of possible functional divergence in both D49 PLA 2 and K49 PLA 2 suggests that there is a pressing need to study structure-function relationship of snake venom group Ⅱ PLA 2.
