摘　　要： 目的用噬菌体肽库技术筛选与β1肾上腺素能(β1-AR)受体有结合活性的小肽,为初步建立一种检测受体数目的方法和研究模拟小肽的生物学活性打下基础。方法以合成β1-AR细胞膜外第2环多肽序列为靶分子,对噬菌体随机12肽库进行亲和筛选,并用酶联免疫吸附分析法(ELISA)和竞争抑制实验鉴定阳性克隆的结合性和特异性。结果经3轮筛选,阳性克隆得到富集,在30个阳性克隆中,有2个阳性克隆可与靶分子有较强结合活性,其中1个能特异性阻断抗β1-AR兔血清多抗与靶分子结合。结论该小肽与靶分子有特异性结合。 Objective To screen peptide ligands of β1-adrenergic receptor (β1-AR) from phage-displayed peptide library. Methods The synthetic peptide of the second extralleular loop of human β1-AR was used to screen the library with affinity selective method. Positive clones was identified with the methods of enzyme linked immuno-sorbent assay and competitive inhibition test. Results After three round washes, two positive phage clones were selected among 30 clones, in which one clone can interrupt purified polyclonal antibody serum against β1-AR binding to β1-AR. Conclusion The peptide ligand can bind to the target specifically.
