摘 要: [目的]调查SE33位点在广东汉族群体的扩增片段长度多态性。[方法]应用荧光标记的引物对SE33位点进行PCR扩增,采用Prism ABI377 DNA自动测序仪分离PCR产物,结合等位基因梯阶和分子量内标进行基因分型。[结果]在114名广东汉族无关个体中观察到了35个等位基因,87种基因型。个体识别能力(DP)值为0.986 1,杂合度(H)为0.912 3。[结论]荧光标记SE33扩增体系个体特异性强,识别力高,是个体识别和亲子鉴定的高效的检测体系。 [Objective]The allele frequency distribution of a complex STR locus SE33 in Guangdong Han population was investigated. [Methods] Analysis was carried out using the fluorescent labeled primer and Prism ABI377 DNA sequencer and genotyping was made by comparing with allelic specific ladder and the internal lane standard. [Results]35 alleles and 87 genetypes were observed in 114 unrelated individuals. The power of discrimination and the heterozygosity value were 0. 986 1 and 0. 912 3, respectively. [Conclusion]SE33 locus is a highly informative and polymorphic STR and suitable for individual identification and paternity testing.
