摘 要: 为了提取芒果子叶总RNA ,去除次生物质和多糖等杂质的严重干扰 ,采用了正交试验快速优化提取方法。实验证明 70 %的丙酮能有效地去除大部分杂质 ,提取过程一天内可完成 ,RNA含量可达 67 7μg g·FW ,A2 60 A2 80 比值为 1 95 ,RNA的纯度及质量适合后续反应。 Isolation of high quality RNA from mango cotyledon is difficult because the tissue is rich in secondary substances and carbohydrates combined with RNA. An orthogonal array design for optimizing RNA extraction from cultural cotyledon was described. Seventy percent of acetone was very effect to remove secondary substance in the cotyledons, and the procedure can be completed within one day. RNA yields gave 67 7μg per gram FW with an average A 260 /A 280 of 1 95, and this highly stable, intact and sufficiently pure RNA was suitable for cDNA synthesis.
