摘 要: 由绿色木霉(Trichoderma viride)A10菌株的粗酶制剂中分离纯化得到 6个β-葡萄糖苷酶组分,对它们的物理、化学性质和对20种糖苷类化合物的水解作用进行了测试。其中5个组分都是既能水解β-构型的糖苷化合物,又能水解α-构型的糖苷化合物,表明它们对α-、β-异头专一性的要求并不象文献中报导的那样十分严格,对糖基和糖苷配基专一性的要求也并不十分严格。由此,对葡萄糖苷酶类的命名分类方法提出了讨论,并对在纤维素酶系研究中纤维二糖酶活力的检测方法进行了讨论。 Six b-glucosidase components from T.viride A 10-an excellent cellulase-producting fungus have been separated and purified to homogenous stage by DEAE-and CM-ion exchange chromatography.Their physical and chemical properties, PI, molecular weight, glycosyl content and absorption coefficient were determined.The amino acid analysis of purified components showed that the enzymes have a high content of acidic and hydrophobic group.Their substrate specificity toward 20 glycoside compounds were compared.The results suggested that most of these components can act on a wide range of glucosides(**********a or ********b glycosidic linkage, variety of glycones and aglycones) For example, five enzyme components showed activity toward some ************b-glu-cosides as well as **********a-glucosides, however, only two components can hydrolyze cellob-iose-a main product and inhibitor during cellulose hydrolysis.Nomenclature and classification of the glucosidase classification and the estimating assay of cellobiase activity weve discussed.
