摘 要: 为了快速检测特异性染色体,采用引物原位标记和细胞化学技术,以7号,17号,X和Y染色体特异性寡核苷酸作引物,对外周血淋巴细胞分裂中期染色体和间期核进行定位研究。结果显示,上述4种染色体均得到特异性标记,且信号强,清晰,无背景着色。结论表明,在与染色体有关的许多研究中该法可替代原位杂交,结合其最近在组织病理学中的应用研究,提示该技术有可能成为分子病理学研究和细胞基因快速诊断的有价值的工具。 In order to identify specific chromosomes rapidly, with specific primers for chromosome 7, 17, X and Y, both metaphase and interphase nuclei of peripheral blood lymphocytes were detected by primed in situ (PRINS) labeling technique. Above four chromosomes were successfully labeled with strong and clear signals as well as nonbackground problems. Application of this technique in histopathology suggested that the PRINS technique has the potential to replace conventional in situ hybridization in circumstances for molecular pathology and cytogenetic diagnosis.
