摘 要: 磨皮粉是红莲子加工时机械磨皮的副产物,不仅产量很大,而且营养成分丰富,若不综合利用会导致严重的资源浪费。通过Plackett-Burman试验→最陡爬坡试验→响应面试验的模式设计方法,考察温度、时间等因素对磨皮粉蛋白质提取率的影响,获得磨皮粉蛋白质提取的最优条件是Na2CO3-Na HCO3缓冲液浓度0.03 mol/L、液固比17.5∶1(m L/g)、pH 10.5、提取温度20℃、提取时间1 h、搅拌间隔时间10 min、超声时间7.5 min,此时蛋白质提取率达93.38%。 A large amount of red lotus peel waste, which contains abundant nutrients, is produced during mechanical peeling of red lotus seeds. Accordingly, insufficient utilization of this byproduct is a serious waste of resource. The present study examined the impact of various factors on the extraction efficiency of protein from lotus seed peel waste by the combined use of Plackett-Burman design, steepest ascent design and response surface methodology. We found that 0.03 mol/L Na2CO3-Na HCO3 buffer solution at pH 10.5 was the best solvent for the extraction of lotus seed peel waste protein. A solvent-to-solid ratio of 17.5:1, an extraction temperature of 20 ℃ and 1 h extraction proved optimal, resulting in an extraction yield of 93.38%.
