3种因素对梅菜试管苗玻璃化的影响

摘  要: 研究了以实生幼苗组织为外植体来源时萌发培养基质、外植体的取材部位、培养容器内的空气环境条件对梅菜(BrassicajunceaCoss.var.spnov)试管苗玻璃化的影响。结果表明:①以实生苗下胚轴为外植体,其再生植株中玻璃化苗的比例与实生苗的萌发培养基质中激素(6-BA+NAA)的含量关系密切,激素浓度越高,玻璃苗的比例就越大。②以无激素1/2MS固体培养基上萌发的实生苗切段为培养材料,下胚轴再生植株的玻璃苗发生率最高,茎尖的次之,子叶的最低。③培养容器封口材料的透气性能、继代间隔时间均直接影响着容器内的空气环境条件,故再生植株的玻璃苗发生率与封口材料的透气性能呈负相关关系,而与继代间隔时间呈正相关关系。梅菜玻璃苗的叶绿素含量比正常苗低,有明显的失绿现象。 Effects of three factors:differently-sprouted seedlings,different parts of seedlings and ventilation condition on vitrification of Brassica juncea Coss.var.sp nov regenerated by tis sue culture were studied.The explants were from Brassica juncea Coss.var.sp nov sprouted seedlings.Results indicated that explants from hormone-treated seeds had higher rate of vitrified shoots than those from non hormone-treated seeds.Explants from seedlings sprouted in soil had lower vitrification rate than those from seedlings sprouted in the culture container.Hypocotyl ex plants from seedlings sprouted in the culture container had the highest rate of vitrified shoots,followed by stem apex explants,and cotyledon explants had the lowest.Sealing material and break length of subculture were related to the ventilation condition in the culture container,thus influ encing the vitrification.The vitreous shoots lost green obviously.
