草鱼肠道对10种必需氨基酸的跨壁运输量

摘  要: 采用离体灌注实验系统和茚三酮对氨基酸显色的实验方法,对跨过草鱼肠道壁的氨基酸进行定量分析,在相同的实验环境下,分别研究草鱼肠道对10种必需氨基酸的吸收转运量。结果表明,在60min内草鱼肠道可对灌流的氨基酸进行持续的吸收转运,并在肠道外积累;当肠道内灌流氨基酸浓度逐渐增加时,肠道外只有苏氨酸的浓度与其起始浓度呈正相关变化,其他9种氨基酸在高浓度(10.0mmol/L)时的吸收转运量均显著下降,表现出高浓度氨基酸对吸收转运的"抑制"效应;通过对吸收转运量达到最大值时实验氨基酸的浓度与吸收转运量的比较,以及在氨基酸吸收转运量随时间的变化规律等的比较分析表明,10种氨基酸在肠道吸收、转运量的变化行为特征有一些共性,也有氨基酸种类特异性。总体表现为,草鱼肠道对苏氨酸、异亮氨酸和亮氨酸等具有很高的吸收转运能力,而对精氨酸、苯丙氨酸、组氨酸等的吸收、转运能力较差。 The fingerling Ctenopharyngodon idellus TBwere employed and after 24 h starvation the intestine gut was obtained and cultured in vitro. The ten essential amino cids used in this study were all L-amino acids and the concentration levels were designed at 2.0, 2.5, 5.0, 7.5 and 10 mmol/L, but for Arg, the levels were at 0.5, 1.0, 2.5, 5.0 and 10.0 mmol/L. In each solution, the penicillin and sulfate streptomycin were added in, both at 150 U/mL. The amino acid solution was controlled flowing through the intestine gut at 12-15 drops/min. The concentrations of the ten amino acids in the exudation outside the intestine gut mural was analyses within 60 min and the results showed that the intestine gut absorbed and transferred the amino acids inside the gut continuously and accumulated the amino acids progressively outside the gut during the 60 min. The amino acid concentration in the exudation outside reaching the highest value appeared in the designed levels of 1.0 mmol/L for Arg, 2.5 mmol/L for Val, Leu and His, 5.0mmol/L for Ile and Trp, 7.5mmol/L for Lys, Phe and Thr, and 10.0 mmol/L for Met, respectively. All those indicate that the intestine gut of C.idellus has high ability in absorbing and transportating Thr, Ile and Leu, but low ability in Arg, Phe and His, ect.
