摘 要: 在碱性条件下研究了酸性铬兰K(ACBK)与牛血清白蛋白(BSA)结合的光度性质,探讨了碱性条件下蛋白质与染料相互作用的机理.结果表明:在pH=9.91的条件下蛋白质能使ACBK吸光度降低且吸收光谱紫移,分光光度法和平衡透析法的结合符合Pesavento提出的相分配模型.此外,推导了有机小分子在蛋白质微相和水相中的分配比D的计算公式,并讨论了ACBK在两相中的分配情况. The photometric character of the combination of acid chrome blue K (ACBK) with bovine serum albumin (BSA) was studied in alkaline solutions and their interaction mechanism was preliminarily discussed. The results showed that at pH=9.91, BSA can make not only the absorbance of ACBK reduced, but also its absorption spectrum come into violet shift. Meanwhile, both Spectrophotometric Method and Equilibrium Dialysis Method indicated that the combination of the two coincides with the model of phase distribution presented by Pesavento. In addition, the calculating formula for the distribution ratio D of the organic micromolecules that were in protein phase and water phase was deduced. Based on this, the distribution of ACBK between the two phases was discussed.
