摘 要: 植物组织培养操作过程中的经验积累和技术的掌握是很重要的.在实验过程中常常出现下列问题:培养基母液配制时容易出现钼酸钠难溶解、母液在贮存过程中产生沉淀或有结晶析出、培养基高压蒸气灭菌后会出现灭菌不彻底,而常规的无菌操作工作效率较低且容易导致培养基污染.通过改进母液的配制方法、培养基高压蒸气灭菌时采用二次排气法以及在无菌操作过程中使用无菌铝盘等可解决实验过程常出现的问题;采用固体培养有其局限性,而液体浅层静置培养有着明显的优点,值得在生产中试验推广. The accumulation of experience and acquisition of skills are vital in culturing plant tissue. The difficulty in dissolving Sodium Molybdate in compounding mother liquor of culture medium,the sediment and crystallization in the storage of mother liquor,the survival of bacterium after the extermination of them in highpressure steam and the pollution of culture medium caused by the inefficiency of regular aseptic manipulation are the problems to be solved. The improvement in the process of compounding mother liquor,the adoption of twice outlet of air when exterminating bacterium in highpressure steam and the use of aseptic aluminium plate in aseptic manipulation help to tackle the problems mentioned above in doing experiments. Static culture in shallow liquid is superior to that in solid state,which is worth promoting.
