摘 要: 通过卵泡培养,对比Notch信号通路抑制剂L-658,458对初级卵泡和次级卵泡的影响,发现L-658,458抑制了初级卵泡和次级卵泡的发育,初级卵泡卵母细胞发生了凋亡,实时定量PCR和蛋白印记检测发现初级卵泡卵母细胞中P27在mRNA和蛋白水平表达升高,提示:Notch信号通路可能通过调控初级卵泡卵母细胞内p27表达调控卵母细胞存活。 We compared the influence of L-658,458,one inhibitor of Notch signaling,on development of primary follicles and secondary follicles through follicle culture technique.We found that L-658,458 inhibi-ted the development of both primary follicles and secondary follicles.The apoptosis signal was observed in oocytes of primary follicles.Real time PCR and Western Blotting results showed that p27 mRNA and pro-tein in oocyts of primary follicles was increased.These results suggested that Notch signaling possibly af-fected oocyte survival through regulating p27 expression.
