摘 要: 以黄槐(Cassia surattensis Burm.f)幼嫩叶片为材料,分别接种于附加NAA1ppm.2,4-D 1ppm、6-BA 2ppm的不同基本培养基MS、B_5、Nitsch、Blaydes、SS-B-8、PC-L_2诱导愈伤组织,结果表明,MS+NAA1ppm+2,4-D1ppm+6-BA2ppm诱导愈伤组织效果最好。将愈伤组织转移到MS+NAA1ppm+6-BA2ppm的分化培养基上,培养30天后,即可形成大量的芽苗丛。蔗糖浓度的高低影响芽的分化和苗的正常生长,以2%的蔗糖浓度较适宜,切取幼苗转移到MS(无机盐减半)+IBA2ppm的生根培养基上,培养10天后,在幼苗茎基部长根而形成完整植株。愈伤组织经过多次继代培养仍保留增殖和器官分化的能力。 This paper reports plantlet formation from leaves culture of ornamental legumeLargcanther Senna(Cassia surattensis Burm.f).Immature leaves were cultured on different basic medium(MS,B5,Nitsch,Blaydes,SS-B-8,PC-L_2)with NAAlppm.2,4-Dlppm.6-BA2ppm,the best result ofcallus induction was on MS+NAAlppm+2,4-Dlppm+6-BA2ppm.A large number of buds were formedfrom the calli on MS+NAAlppm+6-BA2ppm.The redifferentiation frequency and growth rate of budswere affected by the concentration of sucrose in medium.The plantlet were obtained from rooting in thebase of stem of shoots cultured on MS(mineral salts were reduced by half in it)+BA2ppm.The calli stillkept the capacity of proliferation and organogensis after some generations of subculture.
