黄瓜杂交种子纯度的RAPD鉴定
The Purity Identification of Cucumber(Cucumis sativus L.) Hybrid with RAPD Markers

摘  要: 鉴定和分析了黄瓜品种真实性,并建立了应用于鉴定亲本及其杂交种子的RAPD指纹图谱.从102条引物中筛选出了应用于黄瓜种子纯度鉴定的引物32条:13条偏母型引物,9条偏父型引物,5条互补型引物,2条特异型引物及3条缺失型引物.利用4条引物S293,S287,S297,S300鉴定3份黄瓜种子的纯度,在94粒种子中有父本混杂2粒,母本混杂1粒及6粒其它种子混杂,从而表明该份黄瓜种子的纯度为90 04%.建立了黄瓜早青二号父本T03雄性系、早青二号母本B35雌性系以及其杂交种子的RAPD的指纹图谱. The variety homogeneity was identified and the parental polymorphisms were analyzed. The primers, which were suitable for the purity identification of Cucumber hybrid seeds, were screened and the fingerprints of RAPD were established among the parents and their hybrid seeds in Cucumber. 32 primers were screened from 102 the ones that could be beneficial to the purity identification of Cucumber hybrid seeds. The results showed that there were 13 primers of partial female type, 9 primers of partial male type, 5 primers of complementary type, 2 primers of peculiar type and 3 primers of shortage type. Hence, their percentages were 1274%, 882%, 490%, 196% and 294% respectively. It was found there were 1 female seed, 2 male seeds and 6 false seeds among 94 Cucumber hybrid seeds when the purity of Cucumber hybrid seeds was identified with 4 primers S287, S293, S297 and S300. Therefore, the purity of the Cucumber hybrid seeds was 9004%. The fingerprints of RAPD were established for ZhaoQing No.2 Female SystemT03, ZhaoQing No.2 Male System B35 and their hybrid seeds in Cucumber.
