摘 要: 目的 :研究高功率微波 (highpowermicrowave ,HPM)对心肌组织结构、超微结构和凋亡相关蛋白表达的影响 ,以探讨HPM对心肌损伤的剂量效应关系、基本病变规律和分子病理机制。方法 :5个不同功率密度的HPM辐照大鼠 ,于伤后 1h ,6h ,2 4h ,7d ,14d及 2 8d采取心肌组织 ,通过光镜、电镜观察其病变规律 ,免疫组化检测Bax ,Bcl_2 ,c_Fos和P5 3蛋白的表达。结果 :HPM辐照后心肌纤维早期出现细胞水肿、水变性 ,中后期可见收缩带、透明样变 ,肌纤维断裂 ,灶性坏死。Bax,c_Fos,P5 3表达明显增强 ,Bcl_2表达一过性增强。结论 :HPM可对心肌造成严重损伤 ,损伤存在剂量效应关系。bcl_2 bax,c_fos和p5 3参与了HPM引起的心肌细胞凋亡。 Objective:To study the irradiation dosage_effect relationship, the rule of pathologic changes and the molecular pathologic mechanisms, myocardial histologic structure and ultrastructure changes observed in rats irradiated by high power pulse microwave(HPM). Methods:Rats were irradiated by HPM of five different power densities,and cardiac muscle was taken at 1?h, 6?h and 24?h, and on d?7, d?14 and d?28. Pathologic changes were observed by light and electron microscopy, and the expression of P53, Bcl_2, Bax and c_Fos was investigated by immunohistochemical method. Results: After HPM irradiation,edema and hydropic degeneration of myocardial fibers happened first; then contraction bands, hyaline degeneration, muscle fiber break and focal necrosis appeared. The expression of P53, Bax and c_Fos was enhanced obviously, and Bcl_2 was enhanced transiently.Conclusions:Myocardial structure was severely injured by HPM. The damage showed dosage_effect relationship. bcl_2/bax, p53 and c_fos were involved in myocardial cell apoptosis induced by HPM.
