摘 要: 目的研究脂肪组织来源的多能干细胞(ADMSCs)体外长期培养和外源基因的表达。方法 常规方法培养脂肪组织来源的多能干细胞,细胞免疫化学方法对未分化的细胞进行鉴定。用不同的物质诱导脂肪组织来源的多能干细胞向脂肪细胞和成骨细胞方向分化。用Ad5βgal转染脂肪组织来源的多能干细胞。结果从成年大鼠的脂肪组织中培养出多能干细胞,在体外生长形态类似成纤维细胞,可以维持未分化状态稳定增殖,体外扩增可超过10代。细胞表达波形蛋白。在不同物质的诱导下,干细胞可分化为脂肪细胞和成骨细胞。脂肪组织来源的多能干细胞体外可稳定表达外源基因。结论 由脂肪组织来源的多能干细胞可在体外培养并稳定增殖,能够表达外源基因,有潜力作为自体移植的一种细胞来源和基因治疗的一种载体。 Objective To investigate the long-term culture in vitro and exogenous gene expression of adipose tissue-derived multipotent stem cells. Methods Adipose tissue-derived multipotent stem cells were cultured routinely. The undifferentiated cells were identified by immunocytochemistry and were induced into adipocytes and osteoblasts by different factors. Ad5β gal was applied to transfect the adipose tissue-derived multipotent stem cells. Results A population of multipotent stem cells were isolated from adult rat adipose tissue. They were processed to obtain fibroblast-like population of cells and could be maintained in vitro for an extanded periods with stable population doubling and they were expanded as undifferentiated cells in culture for more than 10 passages. They were stained by anti-vimentin and could be differentiated into adipocytes and osteoblasts by different factors. All of the cells were transfected by Ad5β gal and expressed lac Z gene. Conclusion Adipose tissue-derived multipotent stem cells could be expanede in vitro, capable of expressing exogenous gene. They were the ideal cell sources for gene therapy and autologous cell transplantation.
