不同氮源对三角褐指藻生长和脂类含量的影响
Effects of Nitrogen Sources on the Growth and Lipid Content of Phaeodactylum tricornutum

摘  要: 研究了不同氮源及浓度对三角褐指藻(Phaeodactylum tricornutum)的生长、总脂及脂肪酸组成的影响。结果表明,三角褐指藻在氮源缺乏时生长缓慢,表现出油脂积累的特性。以初始浓度大于3.9 mmol/L的氯化铵作为氮源,可以促进三角褐指藻生物量和油脂含量的同步增长。高浓度的尿素有利于三角褐指藻的生长,但达到8.0 mmol/L时脂肪含量下降。不同浓度硝酸钠都促进三角褐指藻的生长,但不能促进脂肪的积累。脂肪酸组成分析表明,单不饱和脂肪酸占总脂肪酸的比例为45%~50%,受各种氮源变化的影响较小,可为生物柴油提供较好的流动性。氮源的添加有利于二十碳五烯酸(EPA)和多不饱和脂肪酸(PUFAs)的合成,当氯化铵浓度为3.9 mmol/L时,EPA和PUFAs占总脂肪酸的比例达到最大,分别为20.4%和26.7%。 Effects of different nitrogen sources and their concentrations on the growth,total lipids and fatty acids composition of Phaeodactylum tricornutum were studied.The results showed that P.tricornutum growed poorly in cultures without addition of any nitrogen source,but accumulated lipids effectively.The biomass and lipid content of P.tricornutum were promoted simultaneously with more than 3.9 mmol/L of initial ammonium chloride in medium;8.0 mmol/L of urea enhanced the biomass of algae while decreased the lipid content;in the range of tested concentration,sodium nitrate promoted growth of algae and depressed the accumulation of lipid.The fatty acids composition analysis showed that monounsaturated fatty acid(MPUAs) accounted for 45%~50% of the total fatty acids and was less affected by nitrogen sources,thus provided better mobility for biodiesel.The synthesis of C20∶5(n-3)(EPA) and polyunsaturated fatty acid(PUFAs) were increased with addition of nitrogen sources.With 3.9 mmol/L of ammonium chloride,the ratio of EPA and PUFAs to total fatty acids was the highest,20.4% and 26.7%,respectively.
