摘 要: 本试验研究了离心速率对波尔山羊精子的活率、顶体完整率、存活时间、生存指数的影响 ,以期为波尔山羊的推广、利用提供科学依据和有效方法。对波尔山羊精液进行 10 0 0、2 0 0 0、30 0 0、4 0 0 0rpm离心处理 ,用含葡萄糖、柠檬酸钠、EDTA和卵黄的稀释液代替精清 ,在 4℃下保存 ,每隔 12h检测一次精子活率和精子顶体完整率。结果表明 :在这四种离心处理中以 10 0 0rpm处理效果最好 ,生存指数和保存 4 8h后的精子活率显著优于其它离心处理 (P <0 .0 1) ,精子顶体完整率显著高于其它三种离心处理 (P <0 .0 1) ,精子的存活时间达到 90h ,精子顶体完整率、精子活率和生存指数随着离心转数的增加而下降。离心冲洗处理为精子的冻前保存提供了新的试验方法 ,为进一步深入研究精清和稀释液在精液中的作用提供了新的思路。 The effect of centrifugal speed on preservation of Boer goat semen in low temperature was studied in order to study the variation of speed motility ,Acrosome intact rate ,living time and survival index .Boer goat semen was treated at different centrifugal speed ,1000rpm ,2000rpm,3000rpm and 4000rpm was carried .Semined plasma was replaced by dilute liquid containing glucose,the sodium of citric acid, EDTA and egg yellow .The treat semen was preserved at 4℃ .The semen motility and Acrosome intact rate were detected every 12 hours .The result showed that the effect of centrifugal at 1000rpm was the best .The survival index, semen motility preserving for 48 hours and Acrosome intact rate were significant better that of order centrifugal wish the increasing of centrifugal speed (P<0.01).
