具通用性芸薹属SSR标记在菜心RIL群体中的偏分离分析
Segregation Distortion Analysis of Transferable Brassica SSR Marker in RIL Population of Flowering Chinese Cabbage

摘  要: 为了研究菜心的偏分离遗传特性,以菜心材料"四九-19号菜心"和"3T6"杂交得到的F6重组自交系(RIL)群体为材料,利用从133对芸薹属SSR引物中筛选出的40对SSR引物对RIL群体进行基因分型,结果显示重组自交系中具有78个SSR标记多态性位点,其中46个位点定位在建立的含有10个连锁群的菜心遗传图谱中。偏分离分析发现,遗传图谱中有31个位点表现偏分离,占定位于图谱中位点的67.39%。其中偏向母本"四九-19号菜心"的位点12个,占总偏分离位点数的38.71%;偏向父本"3T6"的偏分离位点19个,占总偏离位点数的61.29%。检测到4个偏分离热点区域,分别位于连锁群LG3、LG4、LG5和LG6上,其中3个偏向父本"3T6"、1个偏向双亲,推测配子体选择可能是产生偏分离的因素。 To study the genetic characteristics of segregation distortion in flowering Chinese cabbage, the F6 recombinant inbred line(RIL) population derived from the cross of'Shijiu-19 caixin'×'3T6'was used as plant materials,the genotyping was performed with 40 SSR primer combinations which screened from 133 Brassica SSR primer pairs.78 polymorphic loci were detected among selected SSR primer combinations in RILs, and 46 of these loci were mapped to the genetic map with 10 linkage groups. The result of segregation distortion analysis for the loci in the genetic map showed that 31 loci, account for 67.39% of total loci in the map, showed segregation distortion. Among of them, 12 loci, account for 38.71% of total segregation distortion loci, were skewed to female parent'Shijiu-19caixin'; and 19 loci, account for 61.29% of total segregation distortion loci, were bias to male parent '3T6'. Four segregation distortion regions(SDRs) were detected in the linkage groups of LG3, LG4, LG5 and LG6. Three of them were distorted to male parent '3T6', and one was skewed to both parents. It may suggest that gametic selection in the RIL population of'Shijiu-19 caixin'×'3T6'may be the result of segregation distortion of polymorphic loci.
