甲基丙烯酸环氧丙酯与DNA的结合作用

摘  要: 运用吸收光谱移动法观察到甲基丙烯酸环氧丙酯Glycidyl Mcthacrylate,GMA与哺乳类基因组DNA具有强的结合作用。GMA浓度在0—2.8mmol/L范围内,DNA的吸收光谱随GMA浓度的增加而增加了向长波方向位移,并伴随吸收强度降低。若固定GMA浓度,改变DNA浓度,GMA吸收光谱也出现与上类似现象。实验还表明GMA与DNA的结合键型为共价型。 To study the mutagenicity of the new mutagen, glycidyl methacrylate (GMA). in mam malian cells, the binding between the mutagen and calf thymus DNA in vitro system was investigated. By using absorption spectrum shift method, it was found that the λmax 202 nm of DNA and λmax 208 nm of GMA shifted by more than 15nm toward longer wave length with decreased absorption intensity af ter the incubation at room temperature for 15 minutes or more. The results indicated that the binding between GMA and DNA occured. The binding force of GMA to DNA was strong and not affected by sodium chloride even at high concentration. It was slightly decreased in the presence of 8M urea. These results revealed that an ionic binding or hydrogen-bond binding between GMA and DNA is un likely, so it was considered to be a covalent bond binding.
