摘 要: 以1g/L升汞、2%次氯酸钠、10g/L苯酚、50/L甲醛、20g/L高锰酸钾和1g/L硝酸银等6种消毒剂对苎麻的叶、茎、根进行表面消毒处理,通过平板计数法分别对培养12、24h后苎麻样品分离出的内生菌数进行统计,结果表明:对于苎麻叶、茎和根不同部位的样品,分别以70%乙醇2min+1g/L硝酸银1min、70%乙醇2min+20g/L高锰酸钾2min和70%乙醇2min+2%次氯酸钠2min的消毒处理,内生菌分离效果相对更好;其中,茎部的内生菌生长主要集中在消毒后12~24h;苎麻成熟期各器官内生菌数目由高到低排列依次为叶>茎>根。 Taking leaves, stems, roots of ramie as materials, surface disinfection treatments were conducted by six kinds disinfectants asfollows: 1g/L mercuric chloride, 2% sodium hypochlorite, 10 g/L phenol, 50 g/L formaldehyde, 20 g/L potassium permanganate, and 1g/Lsilver nitrate. The number of endophyte was counted after cultured 12 h, 24 h by plate count method. The results showed that: 70% ethanol2min + 1g/L silver nitrate 1min treatment on ramie leaf surface sterilized endophyte separation better than others; 70% ethanol 2min + 20g/L potassium permanganate separation surface disinfection is better than other treatments for ramie stem endophyte. 70% ethanol 2 min +2% sodium hypochlorite for disinfection program is a good separation on ramie root endophyte. The number of bacteria endophyte duringmature ramie is leaf > stem> root.
