摘 要: 整体原位杂交 (Whole mountinsituhybridization)用于基因表达定位和表达分布模式的研究 ,已经成为一种非常重要的手段 .PCR方法进行探针标记 ,可以获得特异性高 ,片断大小可变的探针 .采用DIG标记 ,检测已知基因noggin在爪蟾胚胎时空分布 ,所得结果与文献报道一致 。 Whole-mount in situ hybridization has become a very important tool for studying localization and distribution pattern of gene expression.Highly specific probes were obtained,the length of which are variable.With DIG labeling,the temporal and spatial expression of noggin,one of known genes,was detected.The results obtained were consistent with those in paper previously reported,suggesting that the DNA probe produced by PCR is suitable for whole-mount in situ hybridization on Xenopus embryos under specific conditions.
