摘 要: 面向生物学科硕士研究生开设细胞电生理实验可弥补高校细胞电生理实验教学领域的空缺,使学生更好掌握细胞电生理理论知识。论文详细阐述了依托于膜片钳实验技术、以大鼠心室肌细胞动作电位为主要研究对象的细胞电生理实验课程的具体实验内容,相关的实验方法和实验结果。记录大鼠心室肌细胞的动作电位、三种主要成分(Na+、K+和Ca2+)离子电流的分离可以使学生更好理解动作电位产生的离子机制和三种主要成分电流的动力学特性。此外,记录和观察BK单通道和全细胞电流特征可以使学生在认识某种特异性离子通道电流特性的同时加深对膜片钳实验技术的理解和掌握。 The experimental courses of cell electrophysiology for biological graduate students can fill the gaps in the field of cell electrophysiologieal experiments, and enable students to better grasp the theoretical knowledge of cell electrophysiology. This paper describes the detailed contents of the experimental courses relying on patch clamp technique and the related experimental methods and results. Recording action potential of rat ventricular myocytes and identifying the three major ion channel components ( Na +, K + and Ca2+ ) allow students to better understand the mechanism of action potential and the dynamic properties of the three main types of ion channel currents. In addition, observation and recording the properties of BK single-channel and whole-cell current allows students to know the characteristics of a specific kind of ion channel and also to deepen their understanding and mastering in patch-clamping experimental technique.
