地塞米松对大鼠腹腔巨噬细胞抗弓形虫感染及细胞因子分泌的影响
Effects of Dexamethasone on Rat Peritoneal Macrophages against Toxoplasma gondii Infection and Cytokine Secretion

摘  要: 为探讨地塞米松(DXM)对大鼠腹腔巨噬细胞抗刚地弓形虫(T.gondii)和细胞因子分泌的影响,将大鼠随机分为2组,即DXM注射组和空白对照组,连续注射6d,再抽取腹腔巨噬细胞后分为4组,即DXM组和DXM+T.gondii感染组;对照组和T.gondii感染组。通过瑞-吉染色观察T.gondii在巨噬细胞内的增殖;并用半定量RT-PCR检测DXM组和对照组的大鼠腹腔巨噬细胞IFN-γ、TNF-α和IL-2mRNA表达情况用ELISA检测4个试验组细胞培养上清液中3种细胞因子的含量。结果显示,在正常大鼠腹腔巨噬细胞内T.gondii不增殖反而减少,而在DXM注射大鼠后巨噬细胞内T.gondii大量增殖;同时与T.gondii感染免疫密切相关的这3种细胞因子mRNA及其蛋白的表达均被DXM明显抑制。试验表明,大鼠腹腔巨噬细胞对T.gondii具有明显的抗性;但DXM又可诱发腹腔巨噬细胞易感T.gondii,这一结果与细胞因子IFN-γ、TNF-α和IL-2表达下调有关。 To explore the effect of dexamethasone(DXM)on rat peritoneal macrophages against Toxoplasma gondii infection and cytokine secretion,the rats were randomly divided into 2groups:DXM injection group and control group for treatment 6days.After peritoneal macrophages were extracted,the rats were subdivided into four groups:DXM group and DMX+T.gondii infection group;control group and T.gondii infection group.The proliferation of T.gondii in rat peritoneal macrophages was analyzed by WrightGiemsa staining.IFN-γ,TNF-αand IL-2mRNA expressions of peritoneal macrophage in DXM group and control group were determined by RT-PCR.Finally,the cytokine concentration in culture supernatants of 4experimental groups were measured by ELISA.The results showed that the proliferation of T.gondii decreased in normal macrophages,but the proliferation of T.gondii was significantly increased in rat peritoneal macrophages treated with DXM.Furthermore,DXM could significantly inhibit the expressions of the cytokine mRNA and protein in rat peritoneal macrophages.These cytokines are closely related with T.gondii immunity.Obviously,the rat peritoneal macrophages had the significant resistance to T.gondii infection.But DXM could induce macrophages susceptible to T.gondii and this was due to inhibit the cytokine expressions.
