摘 要: 目的 :探讨二甲亚砜 (DMSO)在聚合酶链反应 (PCR)中扩增人类载脂蛋白E基因中的作用 方法 :在不同质量分数DMSO的存在下 ,以PCR技术扩增人类载脂蛋白E基因片段 限制性内切酶酶切检测扩增产物 结果 :在质量分数 5%~ 10 %的DMSO下 ,可以成功地扩增到人类载脂蛋白E基因的第 4外显子片段 结论 :在DMSO作用下 ,PCR扩增增强了特异性 Aim: To study effect of dimethyl sulfoxide (DMSO) on polymorphism chain reaction for human apoE gene. Methods: The exon 4 of human Apo E gene was amplified by PCR adding DMSO with different concentration. The PCR products were digested with restriction enzyme. Results: The exon 4 of human Apo E gene was amplified by PCR adding 5%~10% DMSO into reaction solution. The PCR products were digested with HhaI. Conclusion: The specificity for PCR is increased in the presence of DMSO. PCR produts can be used to analysis for restriction fragment length polymorphism. The developed PCR technique with DMSO is a method for screening ApoE gene polymorphism. [
