摘 要: 本研究对西洋杜鹃组培苗进行了愈伤组织芽诱导,并采用高效液相色谱法对培养过程中植物体内ZT、GA3、IAA、ABA四种内源激素进行了测定,以及对比了不同培养基组合对植物体芽诱导效果及内源激素含量的影响。从内源激素的测定结果可以得出ZT的含量对分化的影响有着重要的作用,分化效果随着ZT含量的增加而明显,IAA在分化过程中处在较低水平。而在愈伤组织诱导芽的过程中,GA3的值基本趋于一个稳定的值。低浓度的2,4-D和较高浓度的ZT配合一定浓度的NAA将能有效地促进愈伤组织芽体的分化。 This study is about Rhododendron hybridum callus budding culture. It determined the four endogenous hormones (ZT, GA3, IAA, ABA) by high performance liquid chromatography, and compared the impact of different culture medium on the budding effect and endogenous hormone content. The result shows that the content of ZT has important influence on differentiation function; differentiation increases along with the increase of the content of ZT obviously while IAA at a relatively low level. And in the process of callus bud induction, GA3 value tends to be a stable value. Low concentrations of 2,4-D and high concentrations of ZT with a certain concentration of NAA will be able to effectively promote the differentiation of callus buds.
