油菜开花期QTL图谱整合及开花候选基因挖掘

摘  要: 应用生物信息学手段,从公开发表的文献中收集和整理了439个油菜开花期性状QTL相关标记信息,利用电子定位作图技术,将不同试验中的开花期QTL进行了电子整合,构建了含147个开花期QTL的综合图谱。结果表明,在整合图谱上,开花期QTL在芸薹属A、B和C基因组上呈非均衡分布,主要分布在A1,A2,A3,A10和C2染色体上,同时发现34个可重复检测的开花期QTL位点。将收集到的拟南芥中控制开花的基因通过同源比对分析,挖掘出66个芸薹属开花相关候选基因,并成功将这些基因定位到整合图谱上。本文结果为确定有关油菜开花期性状QTL及芸薹属作物候选基因提供了信息。 The mapping of quantitative trait locus (QTL) is an important foundation of gene map-based cloning or molecular marker-assisted selection in oilseed rape. In the last decades, a mass of QTLs for flowering time trait in oilseed rape has been produced, but the integration and comparison of QTLs from the different mapping populations are less studied because of the lack of common molecular markers. With the release or availability of Brassica rapa and B. oleracea genome sequences, we carried out an in silico integration of 439 quantitative trait loci (QTLs) associated with flowering time trait from 14 different mapping experiments. A total of 147 flowering time QTLs were mapped unevenly on all 19 chromosomes with the most on A3 linkage group in the integration QTL map. A total of 34 reproducible QTLs were indentified and 66 candidate genes for flowering time were revealed by comparative mapping with Arabidopsis genome. These results facilitate the application of QTLs for Brassica research communities, and identification of candidate genes for flowering time in Brassica crops.
