摘 要: 脂尾(臀)性状是绵羊逆境生存之需的必要性状,其在尾臀部位大量囤积脂肪的分子机理仍不明晰。为此,本研究以国外新近报道的X染色体一处可能与尾脂性状关联的SNP为研究对象,检测该位点在我国尾型极端差异的阿勒泰羊、湖羊以及细毛羊群体中的多态性,并验证该位点是否可作为1个有效的分子标记应用于低脂绵羊新品种培育。成功地扩增出了含绵羊X染色体59327581位点SNP的385bp片段,并采用PCR-SSCP方法进行SNP分型,后DNA测序只鉴定出1种基因型。结果表明,我国脂臀型阿勒泰羊、短脂尾型湖羊以及长瘦尾型中国美利奴羊在X染色体59327581位点处无多态性,说明我国绵羊X染色体59327581位点不能作为选育低脂绵羊品种的有效分子标记。 Sheep fat tail(rump)is a necessary trait for survival in difficult times. The mechanism of storage fat in sheep tail and rump is still unclear. Therefore,to take the recently reported SNP which may associate with the trait of fat tail as the research issue in this study,and detect polymorphism of the SNP among three extremely different tail-type sheep breeds(Altay, Hu and Chinese Merino) ,and verify whether the SNP can be used as an effective molecular marker in low-lipid sheep breeding. A 385 bp fragment containing the 59327581th loci SNP on X chromosome was successfully amplified,and PCR-SSCP method was used for SNP genotyping,and only one genotype was identified by SSCP and sequencing. This study indicated that there is no polymorphism of this loci in fat-rump Altay, short fat-tail Hu and long thin-tail Chinese Merino populations, and showed that the 59327581 loci was not suitable as an effective biomarker for selecting Chinese low-fat sheep breeds.
