SIRT1在运动训练和白藜芦醇改善T2DM大鼠骨骼肌胰岛素信号通路中的作用
The Effects of SIRT1 and/or Resveratrol on the Improvement of Insulin Signaling Pathway Obstacles in Skeletal Muscles of Type 2 Diabetic Rats

摘  要: 目的:探讨SIRT1在运动训练和(或)白藜芦醇改善Ⅱ型糖尿病(T2DM)大鼠骨骼肌胰岛素信号通路中的作用。方法:将40只雄性SD大鼠分为正常对照组(C)、T2DM组(D)、T2DM运动组(DE)、T2DM白藜芦醇给药组(DR)和T2DM白藜芦醇给药运动组(DER),每组8只。干预后,检测大鼠空腹及OGTT后30min、60 min和120 min血糖和血胰岛素浓度及比目鱼肌SIRT1、AKT、pAKT-Ser473、IRS-1、pIRS-1-Ser307和PTP1B蛋白表达量。结果:1)与C组比较,D组空腹血糖和空腹血胰岛素明显升高;与D组比较,DE、DR和DER组OGTT后30 min、60 min和120 min血糖浓度明显低于D组对应时间点血糖浓度;与D组比较,DE、DR和DER组空腹血胰岛素浓度明显降低(P<0.01或P<0.05),OGTT后30 min和60 min,血胰岛素浓度明显低于D组对应时间点血胰岛素浓度,120 min仅DER组低于D组浓度(P<0.01)。2)与C组比较,D组SIRT1和pAKT-Ser473明显降低(P<0.01),IRS-1和AKT无明显变化,PTP1B和pIRS-1-Ser307明显升高(P<0.01)。3)与D组比较,DE、DR和DER组AKT和IRS-1无明显变化(P>0.05),SIRT1和pAKT-Ser473显著升高(P<0.01),PTP1B和pIRS-1-Ser307显著降低(P<0.01)。4)与DE和DR组比较,DER组SIRT1和pAKT-Ser473显著升高(P<0.05),PTP1B和pIRS-1-Ser307显著降低(P<0.05)。结论:7周游泳训练和(或)白藜芦醇单独或共同干预均能改善T2DM大鼠糖耐量受损,且共同干预较单独干预效果更加显著。 Objective: It is to observe the effect of SIRT1 and/or resveratrol on the improvement insulin signaling pathway obstacles in skeletal muscles of type 2 diabetic rats (T2DM). Methods: 40 male SD rats were randomly divided into control group (C, n = 8 ),diabetes group ( D, n = 8), diabetes + exercise group ( DE, n = 8), Diabetes + Resveratrol ( DR, n = 8), and diabetes + exercise + resveratrol (DER, n =8) group. The levels of fasting blood glucose and fasting blood insulin and the levels at 30 min, 60 rain, 120 min after OGTr were tested. Also the protein expressions of SIRT1, AKT, pmKT-Ser473, IRS-1, plRS-1-Ser~ and PTP1B were determined in the soleus. Results: 1 ) The concentration of plasma FBG (fasting blood glucose) and FINS (fasting blood insulin) of D group was significantly higher than that of C Group (P 〈0.01 ) ; the concentration of plasma glucose of DE, DR, and DER group at 30 min, 60 min, 120 min after OG'IT was significantly lower than that at the corresponding time for D group ( P 〈 0. 01 ). Compared with D group, the concentration of FINS and that at 30 min, 60 min after OG'IT for DE, DR and DET group decreased significantly (P 〈0. 01 or P 〈0.05 ), while at 120 min after OGTT, only the concentration of blood insulin of DER group decreased significantly ( P 〈0. 01 ). 2 ) The content of SIRT1 and pAKT-Ser^473 in the soleus of D group decreased significantly (P 〈0.01 ), but that of IRS-1 and AKT didn't change obviously (P 〈0.05), and that of PTP1B and plRS-1-Ser^307 increased obviously ( P 〈 0. 01 ). 3 ) Compared with D group, the expressions of AKT and IRS-1 in the soleus of DE group didn' t change significantly, the same as DR and DER group. While that of SIRT1 and pAKT-Ser^473 increased significantly (P 〈0. 01 ), and that of plRS-1-Ser^307 decreased significantly ( P 〈 0. 01 ). 4 ) Compared with DE and DR group, the content of SIRT1 and pAKT-Ser473 increased significantly (P 〈 0.0
