摘 要: 采用实时荧光定量PCR分析中外猪种各11个组织mRNA的差异表达,为进一步研究该基因差异表达的机制,根据人、鼠类胰岛素生长因子1受体(IGF-1R)基因的5'调控区的保守序列设计引物扩增猪的5'调控区序列,并利用PCR-直接测序方法分析大花白及长大二元杂猪种的5'调控区的遗传多态性;生物信息学分析5'调控区的CpG岛。结果表明猪IGF1-R的mRNA表达量在中外猪种的心脏中表达量均为最高,其次是肾脏,在肝脏的表达量差异不显著,在长大二元杂猪的心、肺、背肌、大脑、小脑、垂体表达量均极显著高于大花白猪,分别为6.06、52.56、7.70、2.43、3.63和41.38倍,而在胃、脾、肺的表达量极显著低于大花白猪;获得了1 355 bp的IGF1-R 5'调控区序列,测序结果表明该区域在两猪种当中未发现遗传变异;CpG岛预测发现5'调控区存在长度分别是353 bp、603 bp及264 bp的3个CpG岛。 An analysis of differential mRNA expression levels in respective eleven tissues of exotic and lo- cal pig breeds were carried out by using Real - time PCR. To further study the differential expression mecha- nism of the gene,the 5'regulatory region fragment of IGF - 1 designed based on the conserved sequences of the 5'regulato gene was amplified by PCR and the primers were ry regions in human being and rodent, and genetic polymorphisms between Landrace x Yorkshire and Dahuabai pigs were studied by PCR -direct sequencing technology. Then,three CpG islands in the 5 'regulatory region were predicted by using bioinformatics. The re- suits showed that there was the highest expression levels in hearts in both of the two breeds and no differences in livers between the two breeds. The expression of IGF - 1R mRNA were significantly higher in heart, lungs, M. longissimus, brain and cerebellum, pituitary in Landrace x Yorkshire cross than that in Dahuabai, and the folds of its expression levels between the two breeds were 6.06,52. :56,7.70,2.43,3.63 and 41.38 ,respective- ly. However, its expression in the stomach, spleen,lung, expression was significantly lower than that in. Dahuabai.The 1 356 bp 5' regulatory region sequence was obtained and there were not any polymorphisms in this region in the two pig breeds. Three CpG islands ,which lengths respective 353 bp ,603 bp and 264 bp ,were found in the 5'regulatory region.
